Low oxygen tension affects proliferation and senescence of caprine bone marrow mesenchymal stem cells in in vitro culture condition

The culture system of bone marrow mesenchymal stem cells (bmMSCs) in the normoxic environment does not imitate hypoxic milieu typical biological conditions, thus conditions may improve survival, and growth attributes bmMSCs during vitro culture. Therefore, present study was conducted at ICAR-CIRG, Makhdoom year 2020 with objective to investigate changes characteristics cultured caprine (cbmMSCs) including cellular senescence, rate proliferation, immuno-phenotypic characteristics, gene expression pattern a normal microenvironment condition. For this, cbmMSCs isolated from collected iliac crest were enriched grown under either (5% O2) or (20% conditions. Thereafter, outcome culturing on profile important stemness-associated (OCT-4) oxidative stress [glutathione peroxidase (GPx1) copper-zinc superoxide dismutase (CuZnSOD)] marker genes evaluated. cultivated showed higher proliferation decreased population doubling time senescence-associated β-GAL expression; however, immune-phenotypic remain unchanged. Furthermore, hypoxia increased OCT-4, GPx1, CuZnSOD, compared normoxia. In conclusion, condition low O2 level improved cbmMSCs. These outcomes would provide information formulate strategies for collection efficient expansion goats other farm animals before their downstream applications.
